Peptide mapping of proteins by capillary electromigration methods.

This review article provides a wide overview of important developments and applications of capillary electromigration methods in the area of peptide mapping of proteins in the period 1997-mid-2022, including review articles on this topic. It deals with all major aspects of peptide mapping by capillary electromigration methods: i) precleavage sample preparation involving purification, preconcentration, denaturation, reduction and alkylation of protein(s) to be analyzed, ii) generation of peptide fragments by off-line or on-line enzymatic and/or chemical cleavage of protein(s), iii) postcleavage preparation of the generated peptide mixture for capillary electromigration separation, iv) separation of the complex peptide mixtures by one-, two- and multidimensional capillary electromigration methods coupled with mass spectrometry detection, and v) a large application of peptide mapping for variable purposes, such as qualitative analysis of monoclonal antibodies and other protein biopharmaceuticals, monitoring of posttranslational modifications, determination of primary structure and investigation of function of proteins in biochemical and clinical research, characterization of proteins of variable origin as well as for protein and peptide identification in proteomic and peptidomic studies.

Application of Capillary Electromigration Methods in the Analysis of Textile Dyes-Review.

Fiber traces are one of (micro)traces that can be found at a crime scene. They are easily transferable and, like other forms of evidence, can provide a link between a suspect and a victim. The main purpose of this review is to present methods developed to examine textile dyes extracted for forensic purposes using different capillary electromigration methods (CEMs). Scientific papers, mainly from the 20th century, provide reliable methods for the separation of water-soluble dyes. However, dyes insoluble in aqueous solutions have been and still are a challenge. Another problem is the sensitivity of the developed methods, which is, in most cases, insufficient for forensic examination of dyes extracted from a single fiber preserved at the crime scene. Although the methodologies already developed and presented in this review have the potential to be applied in a comparative analysis of textile dye traces, there seems to be a lot of work to be conducted. Some ideas on how to resolve these problems are presented and discussed in the article.

Capillary electromigration methods for food analysis and Foodomics: Advances and applications in the period February 2019-February 2021.

This work presents a revision of the main applications of capillary electromigration methods in food analysis and Foodomics. Articles that were published during the period February 2019-February 2021 are included. The work shows the multiple CE methods that have been developed and applied to analyze different types of molecules in foods. Namely, CE methods have been applied to analyze amino acids, biogenic amines, carbohydrates, chiral compounds, contaminants, DNAs, food additives, heterocyclic amines, lipids, secondary metabolites, peptides, pesticides, phenols, pigments, polyphenols, proteins, residues, toxins, vitamins, small organic and inorganic compounds, as well as other minor compounds. The last results on the use of CE for monitoring food interactions and food processing, including recent microchips developments and new applications of CE in Foodomics, are discussed too. The new procedures of CE to investigate food quality and safety, nutritional value, storage and bioactivity are also included in the present review work.

Recent developments and applications of capillary and microchip electrophoresis in proteomics and peptidomics (2015-mid 2018).

This review summarizes recent developments and applications of capillary and microchip electroseparation methods in proteomic and peptidomic analyses since the year 2015 to ca. mid 2018. Sample preparation procedures for the removal of interfering components or for pre-fractionation and preconcentration of proteins and peptides of interest are discussed. The innovations in coupling of capillary or microchip electroseparation methods with different modes of mass spectrometry detection are covered. In addition, significant recent applications of capillary electromigration methods in both bottom-up and top-down proteomics as well as in determinations of post-translational modifications of proteins are presented. Moreover, several examples of the utilization of capillary electromigration methods coupled with mass spectrometry detection for clinical proteomics and peptidomics are described.

This review work presents and discusses the main applications of capillary electromigration methods in food analysis and Foodomics. Papers that were published during the period February 2015-February 2017 are included following the previous review by Acunha etal. (Electrophoresis 2016, 37, 111-141). The paper shows the large variety of food related molecules that have been analyzed by CE including amino acids, biogenic amines, carbohydrates, chiral compounds, contaminants, DNAs, food additives, heterocyclic amines, lipids, peptides, pesticides, phenols, pigments, polyphenols, proteins, residues, toxins, vitamins, small organic and inorganic compounds, as well as other minor compounds. This work describes the last results on food quality and safety, nutritional value, storage, bioactivity, as well as uses of CE for monitoring food interactions and food processing including recent microchips developments and new applications of CE in Foodomics.

Analysis of antibiotics by CE and CEC and their use as chiral selectors: An update.

Natural, synthetic or semisynthetic antibiotics are highly used to prevent or treat diseases in humans and animals, and to promote animal growth. This fact makes that antibiotics residues or their transformation products may be present in food or in the environment after human or animal excretion. For this reason, it is imperative to develop reliable and sensitive analytical methodologies for their analysis. The main aim of this work is to present and discuss the most recent applications of capillary electromigration methods for the analysis of antibiotics, including the developments and applications of their use as chiral selectors in CE. The literature published from June 2015 to June 2017 is included following the previous review by Domínguez-Vega etal. (Electrophoresis, 2016, 37, 189-211). Information about the use of different detection systems, off-line and on-line strategies to improve sensitivity, and microchip devices for the analysis of antibiotics is provided and properly discussed.

This work includes recent results on food quality and safety, nutritional value, storage, bioactivity, as well as applications of CE for monitoring food processing. The use, among other CE developments, of microchips, CE-MS, and chiral CE in food analysis and Foodomics is also discussed.

Following the previous review by Castro-Puyana et al. (Electrophoresis, 2012, 33, 147–167), the current review covers the papers that were published from February 2011 to February 2013.

Papers that were published from February 2009 to February 2011 are included following the previous review by Herrero et al. (Electrophoresis, 2010, 31, 205-228). The analysis of amino acids, biogenic amines, peptides, proteins, DNAs, carbohydrates, phenols, polyphenols, pigments, toxins, pesticides, vitamins, additives, small organic and inorganic ions and other compounds found in foods and beverages are reviewed, as well as those applications of CE for monitoring food interactions and food processing. The use of microchips, CE-MS and chiral-CE in food analysis is also discussed as well as other current and foreseen trends in this area of research including new developments in Foodomics.

Chiral CE‐MS

This review article addresses the developments and applications of capillary electromigration methods coupled on-line with MS for chiral analysis. The multiple enantiomeric applications of this hyphenated technology are covered including chiral analysis of drugs, food compounds, pesticides, natural metabolites, etc. in different matrices such as plasma, urine, medicines, foods, etc. This work intends to provide an updated overview (including works published till September 2009) on the principal chiral applications carried out by CZE-MS, CEC-MS and MEKC-MS, discussing their main advantages and drawbacks in all their different areas of application as well as their foreseeable development in the not too distant future.

The article gives a comprehensive review on the recent developments in the applications of high-performance capillary electromigration methods, zone electrophoresis, isotachophoresis, isoelectric focusing, affinity electrophoresis, electrokinetic chromatography, and electrochromatography, to analysis, preparation, and physicochemical characterization of peptides. The article presents new approaches to the theoretical description and experimental verification of electromigration behavior of peptides, covers the methodological aspects of capillary electroseparations of peptides, such as rational selection of separation conditions, sample preparation, suppression of peptide adsorption, new developments in individual separation modes, and new designs of detection systems. Several types of applications of capillary electromigration methods to peptide analysis are presented: conventional qualitative and quantitative analysis, purity control, determination in biomatrices, monitoring of chemical and enzymatical reactions and physical changes, amino acid and sequence analysis, and peptide mapping of proteins. Some examples of micropreparative peptide separations are given and capabilities of capillary electromigration techniques to provide important physicochemical characteristics of peptides are demonstrated.

Papers that were published during the period June 2002-June 2005 are included following the previous review by Frazier and Papadopoulou (Electrophoresis 2003, 24, 4095-4105).
